Entecavir (monohydrate) is a drug used to treat hepatitis B virus infection. Entecavir (monohydrate) is a guanine nucleoside analog with inhibitory effect on hepatitis B virus (HBV) polymerase. It is able to become an active triphosphate by phosphorylation, which has an intracellular half-life of 15 hours. Entecavir triphosphate inhibits all three activities of viral polymerase (reverse transcriptase) by competing with deoxyguanine nucleoside triphosphate, the natural substrate of HBV polymerase: (1) initiation of HBV polymerase; (2) the formation of pregenomic mRNA reverse transcription negative strands; (3) Synthesis of HBVDNA positive chain.
