The immunosuppressive mechanism of mizoribine mainly reduces the synthesis of guanylate by inhibiting hypoxanthine nucleotide dehydrogenase (IMPDH) and guanine nucleoside monophosphate synthetase (GMP), reducing RNA and DNA synthesis, thereby inhibiting lymphocyte proliferation and inhibiting the production of antibodies, prolonging the survival time after organ transplantation, and preventing cells from entering the S phase from the G1 phase.
Mizoribine is mainly used to suppress rejection before and after kidney transplantation, and also for liver transplantation and autoimmune diseases. Mizoribine can inhibit the rejection of gastric transplantation, have the pharmacological effect of specifically inhibiting lymphocyte proliferation, inhibit the embryonic transformation reaction caused by various mitotic factors, and prolong the life of transplanted organs. By antagonizing the synthesis process of purine, nucleic acid synthesis is inhibited by the pathway of inosine to uronic acid, but does not penetrate into polymer nucleic acids.
